Vehicles can sometimes be a bitch to line up.
It dosnt matter how many vehicles you have .. one vehicle out of line can cause all vehicles to run off line on that side of the road.
To check, go to  both ends of the map to see if the cars are crossing the centre line, if they are out of line they might start on the correct side but they will cross over by the time they get to the other end of the map.
Most people line up objects manually by simple using the rotate tool. But for vehicles , trains and train tracks a more accurate method is available in the editor.
You can get a true alignment by entering the correct number in the co-ordinate box.
To change the co-ordinates, select the model, double click each number in the PYR box and when it turns blue, simply type in the correct number in each box. (you have to do one at a time) then when all three are reset click the little box at the end of the PYR line.
Your model should flick around to the new alignment.
Depending on which way, north or south the vehicle is facing the co- ordinates shoud be

North/South
P=180 or 0
Y=180  or 0
R= 180 or 0,

Example 1 VW facing south

East/West
P= 180 or 0
Y=90 OR 270
R= 180 0R 0

Make sure the " keep ojects on ground at change" and the ' Align to ground normal' buttons are unchecked or you can get some wierd results.

You can also use this method to line up train tracks and trains, bridges fences etc.

OK KY

Check also to see if you have an extra vehicle that is under the ground or floating above the road. It will pop back onto the road and run in game, but you wont see it in the editor unless you use the 'select all' button on each one of the vehicles and look for an extra green highlighted square.

If that dosnt work..
LOOK for me on TS and I will get you to upload your folder so I can have a look at it for you.
